CAPSI : Chaîne d’Analyse Protéomique multifactorielle haute Sensibilité à composants Intégrés


The CAPSI project (Chaîne d’Analyse Protéomique multifactorielle haute Sensibilité à composants Intégrés; Multifactorial, high sensitivity proteomics process with integrated components) aims to detect and quantify protein biomarkers in blood samples in multiplex assays run on a miniaturised proteomics analysis system. The long-term aim is to develop innovative diagnostic tools, or "health portals", which would allow faster, more reliable biomedical assays in a clinical setting.​

Researcher at our laboratory are specialised in proteomics analysis (analysis of the proteins present in complex biological samples). They developed and optimised a process for the detection and absolute quantification of protein biomarkers in blood samples. This process involves a trypsin digestion step, followed by analysis combining liquid chromatography (LC) and mass spectrometry (MS). Absolute quantification of biomarkers was possible using the PSAQ™ method (Protein Standard Absolute Quantification), where internal standards are added to biological samples. The internal standards are stable isotope labelled protein biomarkers.

The CAPSI project combined the expertise of the EDyP team at our laboratory with that of the Leti/DTBS with a view to developing a miniaturised proteomics process. This project specifically exploits the expertise of the Leti/LCIV to develop microfluidics components, and the Leti/LFCM's know-how for functionalisation of these components. Analytical systems were assessed and optimised by the Leti/LE2S using inverse problem-solving approaches to analyse measurements. The miniaturised system produced consists of various modules, including a protein digestion chip, an LC module and an integrated "nose" for peptide ionisation before MS analysis.
